Three doses of intravenous ketamine, given after detoxification, significantly increased confidence to abstain from alcohol in people with alcohol use disorder. Confidence scores rose 51.98% from baseline after the sessions, though the effect may diminish over time, indicating a need for ongoing support.

A Systematic Review of Methodological Approaches in Indian Studies Evaluating Effectiveness of Ketamine Therapy for Psychiatric Disorders: Recommendations for Future Research

A systematic review of Indian studies on ketamine for psychiatric disorders found that while interest is growing, most of the 13 included studies have significant methodological weaknesses. The studies were mostly single-center prospective trials with small sample sizes (median 25 participants). Only five reported randomization, two blinded outcome assessors, and none used a double-blind design. Adverse effects were typically assessed through unstructured observation or self-report rather than systematic tools. The review recommends that future research adopt rigorous designs including proper randomization, control groups, blinding, structured safety monitoring, and adequate sample sizes to improve reliability and generalizability.

Psychedelics: The New Kid on the Block

Psychedelics, including psilocybin, LSD, and DMT, alter perception, mood, and consciousness by acting on brain 5-HT2A receptors. They reduce between-network synchrony and increase connectivity between brain networks, with effects that may persist for months after a single use. Psychedelics do not produce physical dependence or withdrawal. Clinical trials show high-dose psilocybin reduces depressed mood and anxiety in cancer patients, with 80% of participants maintaining improvements at 6-month follow-up. Ayahuasca shows significant antidepressant effects in treatment-resistant depression compared with placebo. Psilocybin increases abstinence in alcohol-dependent participants. DMT induces rapid altered consciousness with increased global functional connectivity and network disintegration. Research on psychedelics has increased since 1990, with 1,538 publications in 2021.
