Psilocybin, the active compound in 'magic mushrooms,' shows strong therapeutic potential for treating alcohol use disorder (AUD) through psilocybin-assisted psychotherapy (PAP). A literature review of peer-reviewed studies from 2010 onward identified three principal pathways for its effects: neurobiological alterations (such as neuroplastic changes), behavioral and psychological improvements (including increased receptivity to cognitive therapies), and spiritual development (spiritual experiences that improve abstinence). The review supports the use of psilocybin in clinical AUD treatment but notes limitations, such as predisposition to certain mental illnesses. Further research and clinical trials are needed to establish a larger evidence base.

Huntington's disease (HD) is a progressive genetic neurodegenerative disorder with no cure. This study proposes using psilocybin, a compound with neuroprotective, neurotrophic, and neuroplastic effects, to treat neurodegeneration in HD. The experiment will use zQ175 knock-in mice, which model HD, and compare treated and untreated groups. After eight weeks, brain tissue will be analyzed for neuronal death and synaptic density, and motor control will be assessed via rotarod, raised beam, and footprint tests. The expected results are that treated mice will show lower neuronal death, higher synaptic density, and better motor performance than untreated mice. Success could lead to clinical trials for psilocybin in HD.

A systematic review of seven studies found that moderate to high doses of psilocybin therapy reduced depression symptoms in people with major depressive disorder. Single moderate doses (15±5 mg/70 kg) lessened depression, while high doses (25±5 mg/70 kg) produced greater improvements and minimal side effects. A positive link emerged between the quality of the psychedelic experience and symptom reduction, and a dose-response relationship was observed. Secondary outcomes like well-being, rumination, and anxiety also improved. The non-directive therapy approach during high-dose sessions allowed personalized experiences. The review suggests psilocybin is an effective treatment for MDD, though more research is needed.

Psilocybin, a naturally occurring psychedelic compound from Psilocybe fungi, is a serotonin 2A receptor agonist that affects neural plasticity and cognition. The authors propose a study to test whether psilocybin reduces morphine consumption and withdrawal symptoms in morphine-dependent mice, comparing its effects to methadone. They hypothesize that psilocybin will decrease morphine intake in a dose-dependent manner and minimize withdrawal behaviors, potentially offering a cheaper, less dependence-forming alternative for treating opioid addiction. The study uses mouse models with intraperitoneal injection, and future research might explore oral administration.
